I'm Confused....
I have a 2016 1SS Manual transmission. Do we have No Lift shifting? Or not?

Yes, its activated anytime , foot to the floor and making a power shift to the next gear. You cannot lift off the gas during the shift.

Just to be clear, on a stock 6th Gen SS, the throttle has to be 93% or above and the vehicle speed must be at least 31 mph.
