So before i went to get my car a inspection for it to pass emissions i have trifecta tune and I reverted back to stock and when i took it, it didn’t get passed ? If i put it back to the custom tune will that help it pass ?

why did you think the trifecta tune would fail inspection in the first place? The trifecta tune (unless it was super customized) doesn't disable emissions tests or anything...so an emissions test would just run thru it like stock.

When you reset the car ecu, you need a certain amount of time and restarts and miles driven before the emissions tests will pass.

Get a simple bluetooth odb2 dongle and a odb2 app for your phone ..many free ones will have an emissions check test that will let you know when your car is ready to be tested (you've triggered all the necessary things so an emissions test can be done).
