The drive needs to be bus powered and low voltage. So something with either a SSD or 4200rpm 2.5" drive inside should meet the requirements. The PDIM in the 6th gen likely only has USB 2.0 ports, but are noticeably faster than the USB 1.1 in the 5th gen. Any portable HDD that works on a laptop should work on the PDIM port.

If you want USB music to fire up right away on startup, I recommend a drive that has a SSD, as a cold starting platter drive might have some initial lag on power up which could cause the MyLink to time out waiting for it and switch to FM or pop the "connection error" message like it does with Pandora sometimes.

People were able to get 128gb working on the 5th gen, so the limit on the 6th gen could be 256gb or maybe higher. With this info handy, that makes the Gen5DIY media mod kind of worth it. You could bring a bunch of videos with you to play on roadtrips. Screw song playlists, do music video playlists. =)
