Doesn't really matter. Just wash your nice MF towels.

I don't think there's a need to use a separate towel for each section of the car. probably overkill. A more effective method is to get two buckets. One for clean soapy water, and one to rinse the "just used" dirty towels/sponges before reapplying. That way you grab the soapy water from one bucket, wipe down a part of the car, and rinse off the dirt in the second bucket to prevent junk from mixing in with the soap.

And I usually use two 16x16 MF towels when doing a waterless wash...but others may use more. I'm not entering my car in shows...I just want it to look great.
