Fuel, Air, Spark, and Compression must all be correct to avoid a misfire.

Air is unlikely, but the other 3 are all suspect. Swap plugs w/ cyl 3 and inspect the plugs visually for quality of combustion. Then see if the misfire moves from 5 to 3 with the plug, repeat with coil if the plug is fine. And check compression & leak down at some point while the plug is out. If all of that checks out OK, suspect the injector.
